Protein Assays: Understanding Key Methods to Quantify Protein Concentration

Protein assays are analytical procedures used to detect and measure the concentration of proteins in solutions. Accurately determining protein concentration is essential for many areas of research and clinical testing. 

Bradford Assay

The Bradford assay is one of the most popular methods for rapid protein quantification. Developed in 1976 by Marion Bradford, it is based on observing the absorbance maximum shift of Coomassie Brilliant Blue G-250 dye when binding to protein molecules. In its native state, the dye has an absorbance at 470nm which shifts to 595nm upon binding proteins. A standard curve is generated using bovine serum albumin (BSA) to determine protein concentration of unknown samples. Some advantages are its simplicity, speed, and compatibility with detergents and reducing agents. However, it has relatively low sensitivity compared to other assays and is not well suited for membranes or glycoproteins since the dye binds non-specifically.

Bicinchoninic Acid (BCA) Assay

Another Protein Assays used widely is the BCA assay. It involves the chelation of cupric ions (Cu2+) by protein peptides under alkaline conditions, yielding a purple-colored reaction product. The BCA/Cu2+ complex interacts specifically with protein backbone peptide bonds rather than individual amino acids for a more accurate result. Sensitivity is greater than Bradford at 0.5-20μg/mL range. The BCA assay can detect membrane and glycoproteins effectively. However, it has a longer incubation time of 30 minutes compared to 5 minutes for Bradford. Interfering substances also need to be controlled.

Lowry Assay

The Lowry assay is a classic chemical assay measuring color change from peptides interacting with the Folin-Ciocalteu reagent, which detects tyrosine and tryptophan amino acid residues. It is a multistep procedure involving protein alkaline treatment, copper chelation of peptide bonds, and color development with Folin reagent containing phosphomolybdic/phosphotungstic acids. While more laborious than Bradford or BCA, it offers high precision and remains a common reference method. Interference from lab buffers and reducing agents is minimized as well. Limitations include nonspecific binding to certain sugars and degradation of disulfide bonds.

Fluorescence-based Protein Assays

Fluorescent probes have also found wide application in protein assays. The fluorescent dye-binding method measures enhancement when binding tryptophan residues in proteins. It provides greater sensitivity down to 1-10 ng/mL range compared to colorimetric assays. A popular commercial kit involves binding a sulfonated coumarin derivative with tryptophan and tyrosine residues under mild conditions. Fluorescence intensity directly correlates with protein amount. While more sensitive, variation between different proteins and interference require normalization controls.

Absorption Spectroscopy Assays

Protein concentration can also be determined by measuring absorbance of aromatic amino acids phenylalanine, tyrosine, and tryptophan at 280nm. Beer's law is applied using the extinction coefficient calculated for a given protein. This A280 assay is highly accurate but requires the amino acid sequence to determine the extinction coefficient. Advantages include simplicity without need of separate reagents. Drawbacks are inability to distinguish protein from other macromolecules and interference at lower concentrations below 1 mg/mL.
